Simultaneous co-Saccharification and Fermentation of Sago Hampas for Bioethanol Production CIGR Journal
Vincent, Micky A.
Abundance of lignocellulosic biomass provides a good solution to the demands of energy crops in producing biofuel like biodiesel and bioethanol. In this study, bioethanol was produced from sago hampas via the Simultaneous co-Saccharification and Fermentation (Sc-SF) process, at 2.5% and 5.0% (w/v) solid loadings. The processing step in Sc-SF is virtually similar to that of Simultaneous Saccharification and Fermentation (SSF). However, during Sc-SF, two enzymes, amylase and cellulase, were added for the co-saccharification of sago starch and fiber. In addition, Saccharomyces cerevisiae was used to ferment the sugars in the hydrolysates. The Sc-SF samples were analyzed for carbohydrate residues, ethanol and acetic acid using the dinitrosalicylic (DNS) acid...

Sortin2 enhances endocytic trafficking towards the vacuole in Saccharomyces cerevisiae Biol. Res.
Vásquez-Soto,Beatriz; Manríquez,Nicolás; Cruz-Amaya,Mirna; Zouhar,Jan; Raikhel,Natasha V; Norambuena,Lorena.
BACKGROUND: A highly regulated trafficking of cargo vesicles in eukaryotes performs protein delivery to a variety of cellular compartments of endomembrane system. The two main routes, the secretory and the endocytic pathways have pivotal functions in uni- and multi-cellular organisms. Protein delivery and targeting includes cargo recognition, vesicle formation and fusion. Developing new tools to modulate protein trafficking allows better understanding the endomembrane system mechanisms and their regulation. The compound Sortin2 has been described as a protein trafficking modulator affecting targeting of the vacuolar protein carboxypeptidase Y (CPY), triggering its secretion in Saccharomyces cerevisiae. RESULTS: A reverse chemical-genetics approach was used...

Study of a region on yeast chromosome XIII that complements pet G199 mutants (COX7) and carries a new non-essential gene BJMBR
Nobrega,M.P.; Graminha,M.A.S.; Troitskaya,E.N.; Nobrega,F.G..
The mutants of Saccharomyces cerevisiae assigned to complementation group G199 are deficient in mitochondrial respiration and lack a functional cytochrome oxidase complex. Recombinant plasmids capable of restoring respiration were cloned by transformation of mutants of this group with a yeast genomic library. Sequencing indicated that a 2.1-kb subclone encompasses the very end (last 11 amino acids) of the PET111 gene, the COX7 gene and a new gene (YMR255W) of unknown function that potentially codes for a polypeptide of 188 amino acids (about 21.5 kDa) without significant homology to any known protein. We have shown that the respiratory defect corresponding to group G199 is complemented by plasmids carrying only the COX7 gene. The gene YMR255W was...

The correlation between the central carbon metabolic flux distribution and the number of shared enzyme regulators in Saccharomyces cerevisiae BABT
Zhou,Xiangfei; Liu,Lunxian; Shang,Chuanyu; Xu,Haifeng; Ding,Chao; Liu,Qian; Yi,Yin.
The central carbon metabolic system is the upstream energy source for microbial fermentation. In addition, it is a master switch for increasing the production of metabolites and an important part of the microbial metabolic network. Investigation into the relationship between genes, environmental factors, and metabolic networks is a main focus of systems biology, which significantly impacts research in biochemistry, metabolic engineering, and synthetic biology. To this end, the central carbon metabolic flux under a variety of growth conditions or using strains with various genetic modifications was previously measured in Saccharomyces cerevisiae using 13C tracer technology. However, the measured values were not integrated and investigated further. In this...

The eukaryotic Pso2/Snm1/Artemis proteins and their function as genomic and cellular caretakers BJMBR
Bonatto,D.; Revers,L.F.; Brendel,M.; Henriques,J.A.P..
DNA double-strand breaks (DSBs) represent a major threat to the genomic stability of eukaryotic cells. DNA repair mechanisms such as non-homologous end joining (NHEJ) are responsible for the maintenance of eukaryotic genomes. Dysfunction of one or more of the many protein complexes that function in NHEJ can lead to sensitivity to DNA damaging agents, apoptosis, genomic instability, and severe combined immunodeficiency. One protein, Pso2p, was shown to participate in the repair of DSBs induced by DNA inter-strand cross-linking (ICL) agents such as cisplatin, nitrogen mustard or photo-activated bi-functional psoralens. The molecular function of Pso2p in DNA repair is unknown, but yeast and mammalian cell line mutants for PSO2 show the same cellular responses...

The impact of dietary dry yeast on lactation curves of primiparous and multiparous Saanen goats Animal Sciences
Gomes, Ludmila Couto; Alcalde, Claudete Regina; Souza, Rodrigo de; Molina, Bruna Susan de Lábio; Lourenço, Daniela Andressa de Lino; Martins, Elias Nunes.
Twenty-four Saanen goats (fifteen multiparous and nine primiparous) during their lactation period were used, from the kidding until the 200th day of lactation; and were distributed in a completely randomized design and in a factorial arrangement (3 diets x 2 parity orders). The diets were composed of either soybean meal (SBM), soybean meal plus inactive dry yeast (SMDY) or inactive dry yeast (DY) used as the protein source. The analyses of milk yield, body weight and dry matter intake (DMI) were made through the Bayesian methodology. For construct the lactation curves were used the Wood’s nonlinear model. The inclusion of inactive dry yeast in the diets reduced the rate of yield increase until the lactation peak for primiparous goats, but for multiparous...

The Role of Tyrosine 207 in the Reaction Catalyzed by Saccharomyces cerevisiae phosphoenolpyruvate carboxykinase Biol. Res.
Andrade,Cherie; Sepulveda,Carolina; Cardemil,Emilio; Jabalquinto,Ana M.
The functional signifcance of tyrosine 207 of Saccharomyces cerevisiae phosphoenolpyruvate carboxykinase was explored by examining the kinetic properties of the Tyr207Leu mutant. The variant enzyme retained the structural characteristics of the wild-type protein as indicated by circular dichroism, intrinsic fuorescence spectroscopy, and gel-exclusion chromatography. Kinetic analyses of the mutated variant showed a 15-fold increase in Km CO2, a 32fold decrease in Vmax, and a 6-fold decrease in Km for phosphoenolpyruvate. These results suggest that the hydroxyl group of Tyr 207 may polarize CO2 and oxaloacetate, thus facilitating the carboxylation/decarboxylation steps.

The uptake of different iron salts by the yeast Saccharomyces cerevisiae BJM
Gaensly,Fernanda; Picheth,Geraldo; Brand,Debora; Bonfim,Tania M.B..
Yeasts can be enriched with microelements, including iron; however, special physicochemical conditions are required to formulate a culture media that promotes both yeast growth and iron uptake. Different iron sources do not affect biomass formation; however, considering efficacy, cost, stability, and compatibility with Saccharomyces cerevisiae metabolism, ferrous sulphate is recommended.

Trehalose accumulation, invertase activity and physiological characteristics of yeasts isolated from 24 h fermentative cycles during the production of artisanal Brazilian cachaça BJM
Pataro,Carla; Guerra,Juliana B.; Gomes,Fátima C.O.; Neves,Maria J.; Pimentel,Patrícia F.; Rosa,Carlos A..
Trehalose accumulation, invertase activity and physiological characteristics of 86 yeast isolates from short fermentative cycles during the production of cachaça in three artisanal distilleries of the State of Minas Gerais were studied. Among these isolates, 70% were able to grow at temperatures between 40 and 42ºC. Only Saccharomyces cerevisiae isolates were able to grow over 40ºC. Lower temperatures (<40ºC) favoured the growth of yeasts such as Candida parapsilosis-like, C. maltosa-like, Kloeckera japonica, S. exiguus and C. bombicola-like. The isolates from all three distilleries were ethanol tolerant, produced invertase, and accumulate trehalose in the presence of glucose. The strains isolated from distillery A presented more resistance to ethanol...
